TITLE: Measuring molecular forces during clathrin-mediated endocytosis

Clathrin-mediated endocytosis consists of the formation of a vesicle out of a flat membrane in eukaryotic cells. Actin dynamics produces the forces required to invaginate the membrane and pinch it off into a vesicle when membrane tension or turgor pressure are high. However, how much force is produced at the molecular level and how the actin meshwork produces forces has remained elusive, because endocytic structures are transient, out of equilibrium, and diffraction limited. In this seminar, I will present new molecular force sensors our lab recently developed to measure forces in the 3 to 20 pN range on individual molecules in vivo. Application of these force sensors to endocytosis demonstrated that forces are collected and redistributed within the endocytic machinery.
